鲫鱼视网膜谷氨酸受体和GABA受体在爪蟾卵母细胞中的表达

我们以两栖类卵母细胞为功能表达系统,通过注射鲫鱼（Ｃａｒａｓｓｉｕｓｃａｒａｓｓｉｕｓ）视网膜ｍＲＮＡ,利用电压箝及药物灌流手段,系统地研究了鲫鱼视网膜内氨基酸受体的类型和特征,结果如下：（１）Ｇｌｕ受体：ＫＡ可以诱发明显的去极化电流,而且Ｄｉａｚｏｘｉｄｅ能增强ＫＡ诱导的反应,这提示鲫鱼视网膜内某些Ｃｌｕ受体是ＡＭＰＡ选择性亚型（ＡＭＰＡ－ｐｒｅｆｅｒｒｉｎｇｓｕｂｔｙｐｅ）。（２）ＣＡＢＡ受体：ＧＡＢＡ能诱发一个快速、光滑的内向电流,绝大部分对ＧＡＢＡ的反应可被ｂｉｃｕｃｕｌｌｉｎｅ所压抑,而ＧＡＢＡ＿Ｂ受体的激动剂ｂａｃｌｏｆｅｎ则无任何作用,这提示,鲫鱼视网膜内大部分是ＧＡＢＡ＿Ａ受体。     

正常小鼠高频心电图时域值和功率谱的研究

本文用南京新博公司生产的ＮＨＥ－１０００型心电高频信息检测分析仪研究了正常小鼠（昆明种）高频心电图（ＨＦ－ＥＣＧ）的时域值和ＱＲＳ波群的功率谱。主要结果如下（以正导为例,－Ｘ±ＳＤ）：心率６０３±８８次／ｍｉｎ（ｎ＝７４）;Ｐ－Ｒ间期相对较长。为３４．９±４．７ｍｓ（ｎ＝５８）,占心动周期的３４．９±４．９％,这与人类有很大的不同;ＱＲＳ波宽９．２±１．２ｍｓ,占心动周期的９．２±１．４％（ｎ＝７４）,这一结果与以前的文献报道相差较大。Ｔ波宽１０．３±３．２ｍｓ,占心动周期的１０．３±３．２％;Ｑ－Ｔ间期１９．４±３．２ｍｓ,占心动周期的１９．５±３．６％;ＱＲＳ波群峰－峰值（Ｖｐ－ｐ）为１．４５６±０．４８０ｍＶ;Ｔ波高０．３３６±０．１１５ｍＶ;７３只动物Ⅱ导联高频切迹总数只有３个,扭挫２６个。Ⅱ导联ＱＲＳ波群的功率谱特点：０—８０Ｈｚ的相对能量为４５．４８±１５．３２％;８０—２００Ｈｚ为４３．９７±９．９５％;２００—３００Ｈｚ为８．８９±７．３８％;３００—１０００Ｈｚ为１．６６±２．７４％。     

反义寡核苷酸片段可抑制鲫鱼视网膜电压依赖性钾离子通道的表达

爪蟾卵母细胞经注射鲫鱼视网总ＲＮＡ后,可表达大量电压依赖性钾离子通道。在此基础上,我们进一步发现,一个特定序列的寡核苷酸能专一地抑制该通道的表达。由于我们设计与合成的该片段与果蝇、小鼠中已克隆的钾通道中编码Ｎ端的一个多肽的ｍＲＮＡ完全互补,因此推测：鲫鱼视网膜中的Ｋ这个区域与其它物种的Ｋ通道有高度同源性,这为克隆该基因和研究它的功能提供了重要资料。

利用苜蓿夜蛾核型多角体病毒（ＡｃＮＰＶ）带β－Ｇａｌａｃｔｏｓｉｄａｓｅ基因标记的非融合蛋白基因转移载体ｐＢｌｕｅＢａｃ将人粒细胞巨噬细胞集落刺激因子（ｈＧＭ－ＣＳＦ）基因成功地插入病毒ＡｃＮＰＶ的基因组中．ｈＧＭ－ＣＳＦ基因在感染重组病毒的草地夜蛾（Ｓｐｏｄｏｐｔｅｒａｆｒｕｇｉｐｅｒｄａ）培养细胞Ｓｆ９中得到表达,感染后的Ｓｆ９细胞培养液能刺激人骨髓细胞在体外形成典型的集落,表达水平可达２．７×１０５５ＣＦＵ／ｍｌ。以ｈＧＭ－ＣＳＦ单抗所作的ＷｅｓｔｅｒｎＢｌｏｔｔｉｎｇ表明,表达的ｈＧＭ－ＣＳＦ对是３种糖基化程度不同的产物,分子量分别约为１５ｋｄ,１８ｋｄ和２０ｋｄ。     

能化态线粒体内膜脂流动性与能量偶联活性

Physical mapping of three fruit ripening genes：Endopolygalacturonase,ACC oxidase and ACC synthase from apple（Malus x domestica） in an apple rootstock A106（Malus sieboldii

The apple rootstock,A106(Malus sieboldii),had 17 bivalents in pollen mother cells at meiotic metaphase 1,and 17 chromosomes in a haploid pollen cell.Karyotypes were prepared from root-tip cells with 2n=34 chromosomes,Seven out of 82 karyotypes(8.5%) showed one pari of satellites at the end of the short arm of chromosome 3.C-bands were shown on 6 pairs of chromosomes 2,4,6,8,14,and 16 near the telomeric regions of short arms.Probes for three ripening-related genes from Malus x domestica:endopolygalacturonase(EPG,0.6kb),ACC oxidase(1.2kb),and ACC synthase(2kb)were hybridized in situ to metaphase chromosomes of A106.Hybridization sites for the EPG gene were observed on the long arm of chromosome 14 in 15 out of 16 replicate spreads and proximal to the centromere of chromosomes 6 and 11.For the ACC oxidase gene,hylridization sites were observed in the telomeric region of the short arm of chromosomes 5 and 11 in 87% and 81% of 16 spreads respectively,proxiaml to the centromere of chromosome 1 in 81% of the spreads,and on the long arm of chromosome 13 in 50% of the spreads. Chromosomal localization of ribosomal DNA sequences in an apple rootstock using a digoxygenin detection system

A 6kb rDNA probe comprising the 18S coding plus spacer sequences has been hybridized to the metaphase chromosomes of apple rootstock cultivar MM106 demonstrating the localization of ribosomal gene arrays in the vicinity of the telomeric regions of the short arms of chromosomes 6 and 14.The in situ results using digoxygenin labelling coupled to an alkaline phosphatase immunoassay were confirmed by silver staining for NORs and nucleoli.This study demonstrates the feasibility of molecular cytogenetic analysis of very small chromosomes(1.0-2.7μm） of apple.
